Alpha channel not working in media file

I have this weird problem I need help resolving.

I have two machines. They are ALMOST identical on purpose.

I created a media file with a lower-third overlay and an alpha channel so that the lower-third can be placed over any camera feed.

The lower-third displays correctly on the lab machine with the camera feed showing behind the lower-third.

I transfer the video lower-third to the production machine, and the lower-third displays a black background and the camera feed is not showing.
It is as if the production machine doesn't recognize that the lower-third clip doesn't have an alpha channel.

I would be grateful for any help with troubleshooting.

Here's what I have: the file starting with Lab is the lab machine, and the file starting with prod is the Production machine. They are both pretty close in specs except the production machine is has a newer motherboard, processor, and more ram. Both even have the same Python config error. :-)
